PRIM2A Inhibitors

PRIM2A inhibitors encompass a range of chemicals that, either directly or indirectly, obstruct DNA replication. Aphidicolin, for instance, specifically targets DNA polymerase α, causing an immediate halt in DNA synthesis. Hydroxyurea functions a step before this, by curtailing the synthesis of deoxyribonucleotides, which are vital building blocks for DNA synthesis.

A considerable faction of the chemicals listed are nucleoside or nucleotide analogs. Gemcitabine, 5-Fluorouracil, Ara-C, Didanosine, and Trifluridine are exemplary representatives of this class. Their mode of action involves incorporation into the DNA strand during replication. While some cause immediate termination of the DNA chain, others introduce mutations or structural changes, leading to replication arrest. Further focusing on DNA topology and structure, Etoposide and Camptothecin target topoisomerases. While Etoposide inhibits topoisomerase II, preventing DNA relaxation, Camptothecin targets topoisomerase I, inducing DNA damage during replication. Mitomycin C and Bleomycin, on the other hand, disrupt DNA directly. Mitomycin C functions as an alkylating agent, forming crosslinks, whereas Bleomycin induces DNA breaks. Lastly, Daunorubicin introduces further complexity by intercalating into DNA, which blocks both replication and transcription.
