PARP-8 Activators

PARP-8 Activators are a specific class of chemicals that directly or indirectly increase the functional activity of PARP-8, a DNA repair enzyme. These activators work primarily by inducing DNA damage or blocking DNA repair mechanisms, enhancing the recruitment and activity of PARP-8 at DNA damage sites. For instance, Olaparib and Rucaparib, which are PARP inhibitors, can enhance the functional activity of PARP-8 by blocking other PARPs, thereby increasing the recruitment of PARP-8 to DNA damage sites. Similarly, Topotecan, Etoposide, and Camptothecinand Bleomycin, Doxorubicin, and Zeocin, which all induce DNA double-strand breaks, can activate PARP-8 by causing DNA damage, thereby enhancing its recruitment and activity at DNA damage sites.

Moreover, chemicals like Cisplatin and MMS, which form DNA adducts or alkylate DNA, respectively, induce DNA damage that can activate PARP-8, enhancing its recruitment and activity at the sites of DNA damage. Hydroxyurea and Aphidicolin induce DNA replication stress, which can also activate PARP-8. The increase in DNA damage or replication stress caused by these chemicals enhances the recruitment and functional activity of PARP-8 at DNA damage sites, thereby providing diverse mechanisms for the functional activation of PARP-8.
