OR6C2 Activators

Chemical activators of OR6C2 can engage in various intracellular signaling pathways to exert their stimulatory effects. Forskolin, for instance, directly stimulates adenylyl cyclase, thereby increasing the levels of cAMP within the cell. The elevated cAMP acts as a secondary messenger that activates protein kinase A (PKA), a kinase that is known to phosphorylate a broad spectrum of target proteins, including G protein-coupled receptors (GPCRs) like OR6C2. This phosphorylation can lead to a change in conformation and function of OR6C2, effectively leading to its activation. Similarly, Isoproterenol, a beta-adrenergic agonist, elevates cAMP levels in the cell, which also leads to the activation of PKA and subsequent phosphorylation and activation of OR6C2.

In addition to pathways that involve cAMP, other signaling molecules and ions also play roles in activating OR6C2. Capsaicin operates by activating the TRPV1 receptor, which is known to influence secondary signaling pathways that can cross-talk with GPCR mechanisms, ultimately leading to the activation of OR6C2. Another example is Zinc gluconate that may interact directly with OR6C2 or other GPCRs and stabilize them in their active conformation. Icilin and Menthol exert their effects through the activation of TRPM8 channels, which results in altered intracellular calcium levels, cascading into the activation of OR6C2. The phosphodiesterase type 5 inhibitors, Vardenafil and Sildenafil, prevent the breakdown of cGMP, which similarly to cAMP can activate protein kinases that can then activate OR6C2. Pilocarpine, by activating muscarinic acetylcholine receptors, leads to an increase in intracellular calcium levels, which can activate calcium-dependent signaling pathways, culminating in the activation of OR6C2. Cinacalcet activates the calcium-sensing receptor (CaSR), which also affects calcium signaling pathways that can lead to the activation of OR6C2. Furthermore, Adenosine by its action on adenosine receptors, and Histamine through histamine receptors, both activate respective GPCRs that can initiate signaling cascades resulting in the activation of OR6C2, due to the interconnected nature of GPCR-mediated signaling pathways within the cell.
