Forskolin is a diterpene that acts by directly stimulating adenylyl cyclase, leading to an increase in cyclic AMP (cAMP) levels within the cell. Elevated cAMP activates protein kinase A (PKA), a pivotal kinase that can phosphorylate Olr390, thus activating it. Isoproterenol mimics this process through its agonistic action on beta-adrenergic receptors, which also culminates in adenylyl cyclase activation and a subsequent rise in cAMP. The activation of PKA by cAMP can lead to the phosphorylation of Olr390, activating the protein. Similarly, Histamine interacts with H2 receptors and Prostaglandin E2 with EP2 and EP4 receptors, both of which trigger adenylyl cyclase activation followed by PKA-mediated phosphorylation of Olr390.
Further, Glucagon, by binding to its respective receptor, activates Olr390 through a PKA-dependent phosphorylation mechanism after raising cAMP levels. IBMX, Vardenafil, and Zaprinast all prolong the action of cAMP by inhibiting phosphodiesterases, enzymes responsible for cAMP degradation, thereby ensuring sustained PKA activation, which is necessary for Olr390 phosphorylation and activation. Anisomycin, while not directly affecting the cAMP pathway, activates stress-activated protein kinases which can phosphorylate specific substrates, including Olr390, leading to its activation within specific signaling pathways.
