Notch Inhibitors

Chemical inhibitors of Notch target the proteolytic process essential for its activation. The gamma-secretase complex is responsible for cleaving the Notch protein at specific sites within the transmembrane domain, a step that is necessary for the subsequent release and activation of the Notch intracellular domain (NICD). Once released, the NICD translocates to the nucleus, where it can regulate the expression of genes involved in cell differentiation, proliferation, and apoptosis. DAPT, LY-411575, RO4929097, MK-0752, PF-03084014 (Nirogacestat), BMS-906024, DBZ (Dibenzazepine), Avagacestat, Begacestat, Semagacestat, MRK-560, and E2012 are all chemicals that inhibit this precise cleavage step. By preventing the cleavage and activation of Notch, these inhibitors effectively halt the signaling cascade that Notch regulates. The inhibition of gamma-secretase activity by these compounds results in the accumulation of the uncleaved Notch receptor, rendering it unable to contribute to intracellular signaling events.The inhibition of Notch signaling by these chemicals is direct and specific to the activity of gamma-secretase on the Notch protein. For example, DAPT and LY-411575 bind to the gamma-secretase complex and modulate its protease activity, thereby preventing the release of NICD from the membrane-bound Notch precursor. DBZ, Avagacestat, and Begacestat, although structurally different, all act to impede the enzymatic activity of gamma-secretase, thereby inhibiting the Notch pathway. Semagacestat, MRK-560, and E2012, through their inhibitory action on gamma-secretase, ensure that Notch remains in its full-length, inactive form on the cell surface. This inhibition of gamma-secretase activity and the subsequent blocking of Notch activation is a key mechanistic action of these chemicals in inhibiting Notch signaling.
