MARCH3 Inhibitors

MARCH3 inhibitors encompass a variety of chemical compounds that modulate the activity of the E3 ubiquitin-protein ligase MARCH3 by targeting the ubiquitination process or the endosomal trafficking pathways regulated by this enzyme. These inhibitors include proteasome inhibitors such as MG132 and Lactacystin, which increase the levels of ubiquitinated proteins within the cell, thereby indirectly impacting the functionality of MARCH3. By preventing the degradation of ubiquitin-conjugated substrates, these inhibitors affect the dynamic equilibrium of the ubiquitination process in which MARCH3 participates. Proteasome inhibition leads to an accumulation of proteins destined for degradation, which can saturate the ubiquitin-proteasome system and affect the normal turnover of proteins that MARCH3 might ubiquitinate. These compounds act by disrupting the cellular mechanisms and pathways necessary for MARCH3's ligase function, influencing the regulation of protein degradation and trafficking that MARCH3 helps mediate.
