Lipase maturation factor 2 (LMF2) is a protein encoded by the LMF2 gene in humans. It plays a crucial role in the proper folding and maturation of lipoprotein lipase (LPL) and hepatic lipase (HL), which are enzymes essential for the metabolism of lipoproteins. LPL is responsible for the hydrolysis of triglycerides in lipoproteins, such as chylomicrons and very-low-density lipoproteins (VLDL), into free fatty acids, which can be taken up by tissues and used for energy production or storage. Similarly, HL facilitates the clearance of triglycerides from intermediate-density lipoproteins (IDL) and high-density lipoproteins (HDL).
LMF2 is located in the endoplasmic reticulum (ER) membrane, where it assists in the post-translational modification of LPL and HL. It is believed to function as part of a larger complex that is necessary for the correct folding of these lipases, which is a prerequisite for their activity. Mutations in the LMF2 gene can lead to a deficiency in functional LPL and HL, resulting in disorders of lipid metabolism such as combined lipase deficiency (CLD). While the exact mechanisms by which LMF2 facilitates the maturation of lipases are not completely understood, it is evident that LMF2 is indispensable for the proper processing and ultimate enzymatic activity of LPL and HL within lipid metabolism.
