Jade-1 Activators

Jade-1 Activators are a class of chemical compounds that can enhance the functional activity of Jade-1, a protein known for its role in enhancing the ubiquitination and degradation of the oncogene β-catenin. Jade-1 is a short-lived protein that is stabilized by the kidney tumor suppressor protein von Hippel-Lindau (VHL). It has histone acetyltransferase (HAT) activity and is associated with chromatin, indicating a role in gene transcription regulation. Jade-1 activators work by either altering the protein degradation pathway, impacting chromatin structure, or affecting the methylation status of DNA.

Some Jade-1 activators, such as Rapamycin, work by influencing the protein degradation pathway. Rapamycin, an mTOR inhibitor, stimulates Jade-1 activity by inhibiting the mTOR pathway, which is known to regulate Jade-1 stabilization. Other Jade-1 activators, including Trichostatin A, Vorinostat (Suberoylanilide Hydroxamic Acid), Sodium Butyrate, Sirtinol, and Nicotinamide, function by impacting chromatin structure. These compounds are histone deacetylase inhibitors that can alter chromatin structure and gene expression, leading to enhanced Jade-1 functional activity, given its role as a histone binding protein. Furthermore, Jade-1 activators such as 5-Azacytidine, Decitabine, and Zebularine function by influencing the methylation status of DNA. These DNA methyltransferase inhibitors can cause DNA hypomethylation, which can lead to increased expression of Jade-1, thereby enhancing its functional activation. In addition, Parthenolide, a NF-kB inhibitor, can disrupt the degradation of Jade-1, leading to its functional activation.
