INTU Inhibitors

INTU (inturned planar cell polarity protein) is a crucial component of the planar cell polarity (PCP) signaling pathway, a conserved signaling cascade that regulates the coordinated orientation of cells within epithelial tissues during development. In vertebrates, INTU is known to interact with other core PCP proteins, such as VANGL and FZD, to mediate cellular processes including cell migration, tissue morphogenesis, and organogenesis. Specifically, INTU plays a role in establishing the polarized distribution of cellular components along the axis perpendicular to the apical-basal axis of epithelial cells. This function is essential for proper tissue organization and patterning during embryonic development, as defects in PCP signaling can lead to a range of developmental abnormalities and congenital disorders.

Inhibition of INTU function can be achieved through various mechanisms aimed at disrupting its interactions with other PCP components or interfering with its signaling activity. One approach to inhibiting INTU is through the use of small molecule inhibitors that target its protein-protein interactions, preventing the formation of functional PCP protein complexes. Additionally, genetic or molecular techniques such as RNA interference or gene editing may be employed to suppress INTU expression levels, thereby reducing its availability for complex formation and downstream signaling. Furthermore, modulation of post-translational modifications or subcellular localization of INTU may also offer avenues for inhibiting its function within the PCP pathway. Overall, elucidating the mechanisms of INTU inhibition provides insights into its role in PCP signaling and its role as a target for interventions aimed at correcting PCP-related developmental defects.
