HSFY2 (Heat Shock Transcription Factor Y-Linked 2) inhibitors operate via a multitude of distinct pathways to specifically affect the stability and function of the HSFY2 protein. The chemicals 17-AAG, Geldanamycin, Celastrol, SNX-2112, and PU-H71 are inhibitors of the heat shock protein 90 (HSP90). They operate by inhibiting the chaperone interaction between HSP90 and HSFY2, which in turn promotes the degradation of HSFY2 through the ubiquitin-proteasome system. Moreover, MG132 directly targets the proteasome to promote degradation by inhibiting the ubiquitin-mediated proteolysis of HSFY2. Nutlin-3 provides another angle of approach by inhibiting MDM2 and thereby elevating p53 levels, which have a down-regulating effect on HSFY2. This multi-pronged approach ensures a comprehensive inhibition of HSFY2 through direct and indirect means.
