They play an essential role in the adaptive immune system by presenting exogenously-derived antigenic peptides to CD4+ T cells. HLA-D is not a single molecule but rather a group of molecules, including HLA-DR, HLA-DP, and HLA-DQ, each with their own set of alleles. These molecules are composed of two chains: an α chain and a β chain, which together form a peptide-binding groove. The peptides presented by HLA-D molecules are typically found in pathogens or other exogenous substances that have been internalized and processed by the antigen-presenting cell. Once loaded with the appropriate peptide, the HLA-D molecule travels to the cell surface, where it can interact with T cell receptors, initiating a series of signaling events that lead to T cell activation and differentiation
