HEATR3 Activators

HEATR3 activators largely operate through two major cellular routes: ribosomal protein assembly and NOD2-mediated NF-kappaB signaling. Emetine, MG132, Leptomycin B, Anisomycin, Resveratrol, Rapamycin, Sulforaphane, and Parthenolide all influence ribosomal protein transport and 5S RNP assembly. The urgency for ribosomal protein replacement or repair indirectly activates HEATR3, as it is integral for the transport and assembly of ribosomal proteins. These chemicals induce a state where the efficiency and fidelity of ribosomal proteins and 5S RNP assembly are compromised, calling HEATR3 into action.

On the other hand, LPS, Curcumin, Piperlongumine, and Rottlerin operate through the NF-kappaB pathway. These chemicals act as activators of NF-kappaB signaling, where HEATR3 has a role through its involvement in NOD2-mediated signaling. By modulating this pathway, these chemicals indirectly activate HEATR3, driving its multifaceted role in both ribosomal function and cell signaling. Activation of NF-kappaB often entails complex signaling cascades, in which HEATR3 acts as an important mediator, hence becoming indirectly activated.
