GPR179 Inhibitors

GPR179 inhibitors like Pertussis Toxin and GDP-beta-S, interact directly with the G-proteins that are essential for transmitting the signals from GPCRs like GPR179 to their intracellular targets. Pertussis Toxin irreversibly inactivates Gi/o-type G-proteins, thereby impeding the receptor's intracellular communication, while GDP-beta-S acts as a competitive antagonist to GTP, preventing the activation of G-proteins.

Others, such as Cholera Toxin, Forskolin, and NF449, exert their effects on the secondary messenger systems that G-proteins regulate. Cholera Toxin and Forskolin influence the cAMP pathway: the former by causing a persistent activation of Gs proteins, and the latter by directly stimulating the production of cAMP, which can have a wide range of effects on cellular processes including those that may counteract GPR179-signaling. NF449 selectively inhibits the Gs alpha subunit, which may disrupt the GPR179-related signaling by undermining Gs protein's function. Further, the list includes compounds like Suramin, YM-254890, U73122, Go 6983, and M119K, each of which targets distinct aspects of G-protein signaling. Suramin acts as a general antagonist to purinergic receptors, which could lead to alterations in GPR179-related signal transduction.
