γ1-Adaptin Inhibitors

The chemical class of γ1-Adaptin inhibitors encompasses compounds that affect the protein indirectly by targeting cellular pathways or processes related to its function. γ1-Adaptin plays a role in clathrin-mediated endocytosis, and thus, inhibitors of this process can serve to alter the functional state of γ1-Adaptin. Compounds such as Dynasore and Chlorpromazine function by hindering the formation of clathrin-coated pits and vesicles, thus affecting the recruitment and function of adaptor proteins including γ1-Adaptin. Wortmannin and Genistein target upstream signaling molecules like PI3K and tyrosine kinases, respectively, altering intracellular signaling cascades that can modulate the activity of proteins involved in vesicular trafficking.

Other compounds, such as Ikarugamycin and Pitstop 2, disrupt the physical interactions necessary for the formation of clathrin-coated vesicles. Brefeldin A and CK-666 target the vesicle formation and actin cytoskeleton rearrangement processes, which are essential for the proper functioning of γ1-Adaptin in vesicular trafficking. Endosidin4, not yet assigned a CAS number, is included based on its action on the exocyst complex involved in vesicle trafficking, which is a process where γ1-Adaptin is also pivotal. Collectively, these inhibitors act on various components of the cellular machinery that are essential for the proper localization and function of γ1-Adaptin, thus serving as indirect means to inhibit its activity.
