FYN Binding Protein 1, often abbreviated as FYB1 or FYB/SLAP-130, is a protein that interacts with the FYN protein, a member of the Src family of tyrosine kinases. Src family kinases are involved in the regulation of numerous biological processes, including cell growth, differentiation, survival, and migration.FYB1 functions as an adaptor protein, meaning it facilitates protein-protein interactions rather than carrying out enzymatic activity itself. It plays a crucial role in the immune response, particularly in T cell activation, by mediating the interaction between FYN and other proteins involved in T cell signaling.

The protein is composed of several domains that enable its interaction with various proteins. Notably, FYB1 contains an SH3 domain, which allows it to bind to proline-rich regions of other proteins, and multiple motifs for binding to Ena/VASP proteins, which are involved in actin cytoskeleton remodeling.In T cells, FYB1 is involved in the formation of the immunological synapse, a specialized contact area between T cells and antigen-presenting cells that allows for efficient communication and signaling. By interacting with both FYN and Ena/VASP proteins, FYB1 can influence cytoskeletal dynamics and signaling events at the immunological synapse.Aberrations in FYB1 functions may contribute to immune-related diseases. For instance, FYB1 has been implicated in autoimmune diseases, with studies showing altered FYB1 expression in conditions like rheumatoid arthritis and systemic lupus erythematosus.
