Phorbol 12-myristate 13-acetate is one such activator which directly stimulates protein kinase C (PKC). Once activated, PKC can phosphorylate FAM130A1, thereby enhancing its activity. Similarly, Forskolin raises the levels of cyclic AMP within the cell, activating protein kinase A (PKA), which can also target FAM130A1 for phosphorylation. Ionomycin, by increasing intracellular calcium concentrations, can activate calcium-dependent kinases that may subsequently phosphorylate and activate FAM130A1. Furthermore, Okadaic Acid and Calyculin A, through their inhibition of protein phosphatases PP1 and PP2A, prevent the dephosphorylation of FAM130A1, maintaining its phosphorylated, active state.

Lastly, Dibutyryl-cAMP, a synthetic analog of cAMP, activates PKA which can lead to the phosphorylation and activation of FAM130A1, expanding the repertoire of pathways that converge on the functional activation of this protein.
