ELP3 Activators

ELP3 activators are a class of chemicals that can indirectly enhance the activity of ELP3, a component of the Elongator complex involved in histone acetylation. The majority of these activators function by inhibiting HDACs or HATs, which are key enzymes in the regulation of histone acetylation and deacetylation, a critical process in the control of gene expression. Inhibition of these enzymes promotes histone acetylation, leading to a more open chromatin state and increased gene transcription. As ELP3 is involved in histone acetylation, these HDAC or HAT inhibitors can indirectly enhance ELP3 activity.

The activator chemicals include HDAC inhibitors such as sodium butyrate, trichostatin A, nicotinamide, curcumin, vorinostat, belinostat, SAHA, tubastatin A, and PCI-34051. Thesecompounds act by inhibiting HDACs to promote histone acetylation, thus indirectly enhancing ELP3 activity. On the other hand, HAT inhibitors such as anacardic acid, garcinol, and C646 work by inhibiting HATs, which can also indirectly activate ELP3. The inhibition of HATs disrupts the balance of histone acetylation and deacetylation, which can indirectly activate ELP3, as ELP3 is part of the Elongator complex involved in histone acetylation.
