DARPP-32 Activators

DARPP-32 is a phosphoprotein that acts as a potent inhibitor of protein phosphatase-1 (PP1) when it gets phosphorylated. The molecule plays an integral role in dopamine signaling, and its phosphorylation state serves as a nexus for integrating various neurotransmitter inputs into dopaminergic neurons. The class of DARPP-32 Activators largely consists of chemicals that activate DARPP-32 indirectly, usually by affecting signaling pathways or cellular processes related to dopamine and cAMP. For instance, forskolin, a known activator of adenylyl cyclase, elevates cAMP levels, which then activate DARPP-32. Compounds like dopamine and its agonists, such as SKF 38393 for D1-like receptors and quinpirole for D2-like receptors, engage in signaling cascades that eventually affect the phosphorylation state of DARPP-32, activating it.

Moreover, certain psychoactive substances like cocaine and amphetamine amplify extracellular dopamine levels, which in turn activate DARPP-32 by stimulating dopamine receptors. Then, there are molecules like phorbol 12-myristate that activates Protein Kinase C (PKC), an enzyme that can phosphorylate DARPP-32. Even antagonists like bicuculline, sulpiride, and spiperone can alter DARPP-32's activity by interfering with neurotransmitter receptors that influence DARPP-32 indirectly.
