CTPS1 Inhibitors

CTPS1 inhibitors encompass a broad spectrum of chemicals that indirectly modulate the activity of CTPS1 by influencing nucleotide metabolism and related cellular pathways. These inhibitors do not bind directly to CTPS1 but affect the enzyme's functionality by altering the availability of substrates or the balance of nucleotides within the cell. For instance, compounds like Hydroxyurea, Fludarabine, and Fluorodeoxyuridine target ribonucleotide reductase, a key enzyme in deoxyribonucleotide synthesis. By affecting the synthesis of deoxyribonucleotides, these compounds indirectly influence the availability of substrates necessary for CTP synthesis, thereby modulating CTPS1 activity.

Similarly, Methotrexate and Leflunomide, by inhibiting enzymes in the folate and pyrimidine synthesis pathways, respectively, create an imbalance in the nucleotide pools. Such imbalances can indirectly impact CTPS1's function, as the enzyme's activity is closely tied to the overall nucleotide metabolism.
