CLS1 Activators

CLS1 activators encompass a group of chemicals that are primarily distinguished by their capacity to indirectly influence the activity of Cardiolipin Synthase 1 (CLS1), a pivotal enzyme involved in cardiolipin synthesis within the inner mitochondrial membrane. A majority of these activators exert their effects by bolstering mitochondrial biogenesis and function, thereby indirectly driving the need for augmented CLS1 activity. For instance, AMPK activators, such as AICAR, are recognized for their role in advancing mitochondrial biogenesis. An escalation in mitochondrial numbers inherently steers a concomitant increase in cardiolipin synthesis requirements, thereby ramping up CLS1 activity.

Further, compounds like Rosiglitazone and Pioglitazone, both PPAR-gamma agonists, bolster mitochondrial biogenesis, which in turn can lead to an uptick in CLS1 activity. Additionally, PGC-1α activators, such as SR-18292, by stimulating PGC-1α, promote mitochondrial biogenesis and therefore can indirectly stimulate CLS1.
