CGI-96 Activators

CGI-96, also referred to as VPS53 or Vacuolar Protein Sorting-associated Protein 53, is a component of the Golgi-associated retrograde protein (GARP) complex, which is involved in the tethering and fusion of transport vesicles to the trans-Golgi network. The corresponding gene that encodes this protein is known as "VPS53."As a part of the GARP complex, CGI-96 plays a crucial role in the retrograde transport of proteins from endosomes to the trans-Golgi network. This retrograde transport is essential for the recycling of proteins, maintenance of organelle identity, and the sorting of lysosomal enzymes. The GARP complex, with CGI-96 as a core component, functions to recognize transport vesicles carrying cargo from endosomes and ensures their specific delivery to the Golgi apparatus.

CGI-96 has also been implicated in the maintenance of cell homeostasis by facilitating the endosome-to-Golgi retrieval of membrane proteins, which is a process vital for the correct localization and functioning of transmembrane receptors and other proteins. Disruptions in this retrieval system can lead to mislocalization and degradation of such proteins, potentially contributing to various cellular dysfunctions.Furthermore, mutations in the VPS53 gene, which would affect the function of CGI-96, have been associated with complex hereditary disorders. For example, research has suggested a link between mutations in VPS53 and neurodegenerative diseases, as well as congenital disorders that affect multiple organ systems, indicating the importance of CGI-96 in a wide range of physiological processes.CGI-96's role in the retrograde transport pathway underscores its significance in maintaining the balance and flow of cellular traffic. Understanding the molecular mechanisms of CGI-96 and the GARP complex provides valuable insights into the regulation of intracellular transport and its implications in human health and disease.
