Centaurin alpha 1 (CENTA1), also known as ADP-ribosylation factor guanine nucleotide-exchange factor 1 (ARFGEF1), is a protein that in humans is encoded by the CENTA1 gene. It is part of the centaurin family of proteins, which are characterized by the presence of a characteristic pleckstrin homology (PH) domain and an Arf-GAP domain.CENTA1 functions as a guanine nucleotide exchange factor (GEF) for ADP ribosylation factors (ARFs), which are small GTP-binding proteins involved in vesicle trafficking and actin remodeling. By activating ARFs, CENTA1 can influence these processes, playing a key role in intracellular trafficking and cellular morphogenesis.
The PH domain of CENTA1 allows it to bind phospholipids, and it's thought to help target the protein to specific cellular locations, such as the plasma membrane. Meanwhile, the Arf-GAP domain provides GTPase activating protein (GAP) activity, turning off ARF signaling.given its involvement in critical cellular processes, it's plausible that dysfunction of CENTA1 could contribute to various pathological conditions. For example, disruptions in vesicle trafficking can contribute to neurological disorders and cancer.
