Cdc42EP3 Activators

Cdc42EP3 Activators are a diverse group of compounds that influence the protein's function through various mechanisms, primarily related to the modulation of intracellular cAMP levels, Rho GTPase signaling, and kinase activity. Cdc42EP3 is a scaffolding protein that interacts with effectors of the Rho family of GTPases, such as Rac1, to modulate their activity. Several of these compounds, such as EGF, LY294002, and Sphingosine-1-Phosphate, mediate their effects through these GTPases, leading to the activation of Cdc42EP3 and enhancement of its scaffolding functions.

Other Cdc42EP3 activators operate through cAMP-dependent signaling and kinase activation. For instance, Epinephrine, Forskolin, IBMX, Caffeine, Prostaglandin E2, and PACAP enhance intracellular levels of cAMP, which activates Protein Kinase A (PKA). PKA can then phosphorylate and activate Cdc42EP3, enhancing its ability to scaffold for effector proteins. Similarly, Bradykinin and Vasopressin operate through the IP3/DAG pathway, leading to Protein Kinase C (PKC) activation. Activated PKC can also phosphorylate and activate Cdc42EP3. LPA, on the other hand, functions through the Rho/Rho kinase pathway, with activated Rho kinase capable of phosphorylating and activating Cdc42EP3.
