CD137 Inhibitors

CD137 inhibitors can be broadly applied to agents that influence the signaling and effects mediated by CD137, a costimulatory molecule expressed predominantly on activated T cells. While the above list doesn't include chemicals that directly bind to and inhibit CD137, they can impede the downstream or associated pathways and processes induced by CD137 activation. For example, Cyclosporine inhibits calcineurin, a pivotal enzyme in T-cell activation. Given the role of CD137 in T-cell activation, the inhibition of calcineurin can indirectly attenuate CD137-mediated effects. Similarly, agents like LY294002 and Wortmannin inhibit PI3K, a kinase with a clear role in T-cell survival and activation downstream of CD137. Another key player in T-cell signaling is the MAPK/ERK pathway, and inhibitors like PD98059 and U0126 can dampen its effects. On the other hand, JNK, influenced by compounds like JNK Inhibitor II and SP600125, is another kinase activated by CD137 and is involved in T-cell activation. Moreover, the importance of NF-kB in T-cell activation is underscored by its inhibitors, BAY 11-7082 and IKK-16. By targeting these essential pathways, the aforementioned chemicals provide avenues to influence the intricate web of signaling induced by CD137.
