CARD 8 Activators

CARD8 activators, such as Thapsigargin and ATP, work by increasing cytosolic calcium levels and activating the P2X7 receptor, respectively, leading to the activation of the NLRP3 inflammasome, which enhances CARD8's activity. Nigericin, Monosodium Urate Crystals, MSU, and Alum also activate the NLRP3 inflammasome by promoting potassium efflux from cells and inducing lysosomal damage, thereby promoting the functional activity of CARD8.

The functional activity of CARD8 is also increased by MDP, which triggers NOD2 and activates the NF-κB pathway. This enhances the inhibitory role of CARD8 on this pathway. Similarly, Amyloid β enhances the activity of CARD8 by causing a NLRP3 inflammasome-dependent inflammatory response. Silica Crystals, like several other compounds, activate the NLRP3 inflammasome and boost the function of CARD8 by inflicting lysosomal damage and promoting the release of cathepsin B. E-64d, a potent cathepsin B inhibitor, indirectly enhances the functional activity of CARD8 by reducing the activation of the NLRP3 inflammasome.

Thapsigargin is an inhibitor of the sarco-/endoplasmic reticulum Ca2+ ATPase (SERCA), causing an increase in cytosolic calcium levels. Since CARD8 is known to be involved in the NLRP3 inflammasome activation which is regulated by intracellular calcium levels, the increase in cytosolic calcium can enhance CARD8’s functional activity.
