Chemical activators of Ly6/PLAUR domain containing 10 can initiate a diverse array of biochemical signals that result in the activation of this protein. Phorbol 12-myristate 13-acetate serves as a potent activator by mimicking diacylglycerol (DAG), which is a physiological activator of Protein Kinase C (PKC). By directly activating PKC, the phosphorylation state of Ly6/PLAUR domain containing 10 can be enhanced, leading to its activation. Similarly, forskolin raises intracellular cAMP levels, which then activate PKA, a kinase that can phosphorylate and consequently activate Ly6/PLAUR domain containing 10. Ionomycin acts by increasing intracellular calcium levels, which subsequently activates calcium-dependent kinases, again promoting the phosphorylation and activation of Ly6/PLAUR domain containing 10.

Epidermal Growth Factor engages its receptor, setting off a cascade that can culminate in kinase signaling pathways that activate Ly6/PLAUR domain containing 10. Thapsigargin contributes to the activation by inhibiting SERCA, causing a rise in cytosolic calcium levels, which, in turn, activate kinases that phosphorylate Ly6/PLAUR domain containing 10.
