ARL4 Activators

ARL4 activators are a group of chemicals that can directly or indirectly enhance the functional activity of ARL4. They influence a variety of signaling pathways and biological processes, significantly impacting the function of ARL4. Phorbol 12-myristate 13-acetate (PMA), for example, is a potent activator of protein kinase C (PKC), which can phosphorylate ARL4, thereby enhancing its GTP binding and function.

Moreover, LY294002 reduces PIP3 levels, enhancing ARL4 function due to reduced PIP3-mediated suppression. Forskolin, an activator of adenylyl cyclase, can increase cAMP levels, which in turn activate PKA that can phosphorylate ARL4, enhancing its function. Calyculin A and Okadaic acid, can enhance ARL4 function by increasing phosphorylation levels. Rapamycin, Metformin, and AICAR, all of which inhibit mTOR, can lead toreduced phosphorylation of 4E-BP1, which can enhance the translation of ARL4 mRNA. Lastly, U0126 and PD98059 can enhance ARL4 function by reducing ERK-mediated inhibitory phosphorylation.
