ARL4 Inhibitors are a group of chemical compounds that primarily work through the modulation of intracellular trafficking processes and the PI3K/Akt signaling pathway. Chemicals that disrupt the structure and function of the Golgi apparatus, such as Brefeldin A, or inhibit the transport of vesicles to the plasma membrane, such as Exo2, can inhibit the functional activity of ARL4, as it relies on these processes for its transportation to the plasma membrane. Additionally, inhibitors of dynamin, such as Dyngo 4a and Dynasore, can disrupt the endocytic pathway that ARL4 is involved in, leading to decreased functional activity of ARL4.
On the other hand, inhibitors of cytohesin and GBF1, such as SecinH3 and Golgicide A, respectively, can disrupt the ARF-dependent endocytic pathway whereARL4 functions, leading to decreased functional activity of ARL4. There are also several compounds, including Wortmannin, LY294002, and Rapamycin, that act as inhibitors of the PI3K/Akt signaling pathway. This pathway is crucial for the membrane localization and functional activity of ARL4. Thus, by inhibiting the PI3K/Akt pathway, these compounds can indirectly inhibit ARL4's function. Similarly, U73122, an inhibitor of phospholipase C (PLC), can affect the functional activity of ARL4, as PLC plays a role in the regulation of ARF GTPases such as ARL4.
