WAP Antibody (E-8) is a mouse monoclonal IgG2a kappa light chain antibody that detects Whey Acidic Protein (WAP) of mouse origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). WAP Antibody (E-8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. WAP is a hormonally-regulated, acidic, cysteine-rich protein primarily found in rodent milk, and plays a crucial role in protease activity regulation, essential for maintaining protein homeostasis during lactation. The WAP gene, consisting of four exons and three introns, encodes two distinct cysteine-rich domains that contribute to structural integrity and functional properties. WAP is expressed in mammary epithelial cells and is significantly induced during pregnancy under the influence of lactogenic hormones such as glucocorticoids, prolactin, and insulin. WAP mRNA expression is tightly regulated and varies with the estrous cycle, being predominantly detected during the midcycle phase. While low levels of WAP RNA can be found in some non-mammary tissues, such as the tongue, pancreas, and pituitary gland, WAP is absent in others like the heart and brain. WAP secretion into milk occurs throughout lactation and is specific to certain species, including mouse, rat, rabbit, camel, and porcine. Mouse mammary epithelial cells cultured on a basement membrane-type matrix express high levels of WAP mRNA and secrete WAP into the lumen, a process dependent on alveoli-like structure formation.
