WAP Antibody (C-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects WAP protein of mouse and rat origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). WAP (C-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Whey acidic protein (WAP) is a hormonally-regulated, acidic, cysteine-rich protein primarily found in rodent milk, and WAP plays a crucial role in regulating protease activity, essential for maintaining milk protein integrity during lactation. WAP gene, consisting of four exons and three introns, encodes two distinct cysteine-rich domains that contribute to WAP′s function as a protease inhibitor. WAP contains a 19-amino acid N-terminal signal peptide that directs secretion. WAP gene expression is significantly induced during pregnancy, driven by lactogenic hormones such as glucocorticoids, prolactin, and insulin, and remains tightly regulated throughout the estrous cycle, with mRNA levels peaking during midcycle. While WAP expression occurs predominantly in mammary epithelial cells, low levels of WAP RNA can be detected in some non-mammary tissues, including the tongue, pancreas, and pituitary gland, but not in the heart or brain. WAP secretion into milk occurs throughout lactation and is specific to several species, including mouse, rat, rabbit, camel, and porcine. Mouse mammary epithelial cells cultured on a basement membrane-type matrix express high levels of WAP mRNA and secrete WAP into the lumen, a process dependent on alveoli-like structure formation.
