VWF Antibody (3E2D10) is a mouse monoclonal IgG1 kappa light chain antibody designed to target von Willebrand factor (VWF), a crucial multimeric glycoprotein integral to the regulation of hemostasis. VWF is synthesized primarily in endothelial cells and megakaryocytes, circulating in the plasma where VWF plays a pivotal role in blood coagulation by mediating platelet adhesion and aggregation at sites of vascular injury. Additionally, VWF serves as a carrier for Factor VIII, a vital cofactor in the coagulation cascade, thereby ensuring efficient clot formation and maintenance of vascular integrity. The functionality of VWF is essential for preventing excessive bleeding, and defects or deficiencies in VWF are responsible for von Willebrand disease, the most common inherited bleeding disorder. Anti-VWF antibody (3E2D10) is optimized for high reactivity with human VWF and is recommended for use in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP), making VWF (3E2D10) antibody a versatile tool for both research and diagnostic applications. By specifically recognizing key domains of VWF involved in platelet binding and Factor VIII interaction, VWF (3E2D10) monoclonal antibody facilitates detailed studies into VWF′s posttranslational modifications, such as propeptide cleavage and intersubunit disulfide bond formation, which are critical for function and stability. Understanding the precise mechanisms of VWF action and VWF′s role in coagulation pathways is essential for advancing treatments for bleeding disorders and thrombotic conditions. Whether employed in basic research to elucidate the molecular underpinnings of hemostasis or in clinical settings to diagnose and monitor VWF-related pathologies, anti-VWF antibody (3E2D10) provides reliable and accurate detection of VWF, supporting advancements in vascular biology and coagulation research.

I am using VWF (3E2D10): sc-21784 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: jerojero
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Rated 5 out of 5 by from strong positive staining of bloods vesselsstrong positive staining of bloods vessels in the upper dermis. The primary antibody could probably be more diluted than 1/500. Similar staining pattern, though slightly weaker, was obtained with 4% formaldehyde fixation.
