VRK1 Antibody (A-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects VRK1 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). VRK1 (A-11) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. VRK1 plays a crucial role in cellular stress responses by phosphorylating key transcription factors, including p53 and ATF2, which are vital for regulating cell cycle and apoptosis. Specifically, VRK1 phosphorylates p53 at Thr18, disrupting p53′s interaction with Mdm2, thereby stabilizing p53 and enhancing tumor suppressor functions. Additionally, VRK1′s interaction with ATF2, where VRK1 phosphorylates Thr73, not only stabilizes ATF2 but also promotes transcriptional activity, which is essential for cellular responses to stress. VRK1′s ability to modulate these critical pathways underscores its importance in cancer biology and cellular homeostasis, making anti-VRK1 antibody (A-11) an invaluable tool for researchers studying these processes.

Do you have a picture/data of use of this antibody Western blot in mouse, or mouse tissues?

Asked by: Effie
Thank you for your question. We do not have data for this antibody with mouse samples. This antibody is not recommended for use with mouse samples due to the low homology between the epitope sequence and the target sequence in mouse.

I used the antibody Sc271061 for Hela cells overexpressing Kinase Dead - VRK1 and the antibody could not recognize the overexpressed protein. Do you know if the antibody was produced against the kinase domain?

Asked by: Robertarrs
Thank you for your question. sc-271061:VRK1 (A-11) was raised against a specific epitope mapping between amino acids 268-297 within an internal region of VRK1 of human origin. If the issue with the experiment persists please contact our technical service for assistance.

What application is the blocking peptide sc-271061 P appropriate for?

Asked by: DefinitelyNotMatt
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization

Rated 5 out of 5 by from excellent for WB but use sensitive substrateI got this 2 weeks ago. It worked very well. Also 3years ago, I published a paper using this antibody. (Oncotarget, 2015, Vaccinia-related kinase 1 promotes hepatocellular carcinoma by controlling the levels of cell cycle regulators associated with G1/S transition)

Rated 4 out of 5 by from Works good with some optimizationI recieved a sample and I was able to have good results after some optimization in time of incubation (mininum overnight) and with a high concentration (dilution 1:250). I always have a non specific band upper VRK1. But its reliable antibody (I tested the specifity with Overexpression and Knockout).

Rated 5 out of 5 by from Excellent antibody for Western blottingThis product has been independently tested and validated in-house at Kinexus Bioinformatics Corporation as a very potent and highly specific antibody for Western blotting with lysates from diverse human cancer cell lines. In immunoblots with a mix of HeLa and Jurkat cell lysates, this antibody very strongly detected a ~46 kDa protein, which corresponds to the predicted size of VRK1.
