V-ATPase B1/2 Antibody (D-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects V-ATPase B1/2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). V-ATPase B1/2 (D-4) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The vacuolar-type H+-ATPase (V-ATPase) is a crucial multisubunit enzyme responsible for the acidification of eukaryotic intracellular organelles, which is vital for processes such as protein degradation, nutrient absorption, and maintaining cellular homeostasis. The V-ATPase operates through a rotary mechanism, consisting of a peripheral V1 domain responsible for ATP hydrolysis and an integral V0 domain that facilitates proton translocation. The V1 domain comprises nine subunits (A-H), while the V0 domain consists of five subunits (a, d, c, c′, and c"). Notably, V-ATPase B1 isoform plays a critical role in the inner ear, where V-ATPase B1 is essential for proton secretion, maintaining the proper pH of endolymph, and ensuring normal auditory function. Mutations in the gene encoding V-ATPase B1 isoform, located on chromosome 2cen-q13, have been linked to distal renal tubular acidosis associated with sensorineural deafness. Meanwhile, V-ATPase B2 isoform, which is expressed in the kidney and osteoclasts, is encoded by a gene mapped to chromosome 8p22-p21. The diverse applications and specificity of V-ATPase B1/2 (D-4) monoclonal antibody make V-ATPase B1/2 (D-4) antibody an invaluable tool for researchers studying the physiological and pathological roles of V-ATPase in various biological contexts.

For Enzymatic antigen retrieval, Can I use Proteinase K?

Thank you for your question. We recommend Pepsin- or Saponin-based enzymatic antigen retrieval: Pepsin: Incubate sections for 10–20 minutes in 0.1% Pepsin (sc-476554) in 0.01 N HCl at room temperature. Wash slides several times in deionized H2O. Aspirate excess liquid from slides. Saponin: Incubate sections for 30 minutes in 0.05% Saponin (sc-280079) in deionized H2O at room temperature. Wash at least three times in PBS. Aspirate excess liquid from slides.
