V-ATPase B1/2 Antibody (F-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects V-ATPase B1/2 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). V-ATPase B1/2 (F-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The vacuolar-type H+-ATPase (V-ATPase) is a crucial multisubunit enzyme that plays a vital role in acidifying eukaryotic intracellular organelles, essential for protein sorting, nutrient uptake, and cellular signaling. V-ATPase operates through a rotary mechanism, consisting of a peripheral V1 domain responsible for ATP hydrolysis and an integral V0 domain that facilitates proton translocation. The V1 domain comprises nine subunits (A-H), while the V0 domain consists of five subunits (a, d, c, c′, and c"). V-ATPase B1 isoform is particularly important in the inner ear, where V-ATPase B1 regulates proton secretion to maintain proper pH of endolymph, ensuring normal auditory function. Mutations in the gene encoding V-ATPase B1 isoform, located on chromosome 2q13.1, have been linked to distal renal tubular acidosis and sensorineural deafness, highlighting V-ATPase B1′s critical role in both renal and auditory health. V-ATPase B2 isoform is predominantly expressed in the kidney and osteoclasts, with V-ATPase B2′s gene mapping to chromosome 8p22-p21, further emphasizing the diverse physiological functions of V-ATPase in different tissues.
