V-ATPase A1 Antibody (E-8) is a mouse monoclonal IgG2b kappa light chain antibody that detects V-ATPase A1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). V-ATPase A1 (E-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. V-ATPase A1 protein plays a crucial role in cellular processes by functioning as a proton pump that regulates pH and ion homeostasis within cells. V-ATPase A1 is primarily located in clathrin-coated vesicles and osteoclasts, where V-ATPase A1 is essential for acidification of intracellular compartments, facilitating processes such as protein degradation and nutrient absorption. V-ATPase A1 consists of two main domains: a hydrophilic amino-terminus rich in charged residues and a hydrophobic carboxy-terminus that features multiple transmembrane regions. This structural arrangement enables coupling of ATP hydrolysis to proton translocation across membranes. Inactivation of osteoclast-specific vacuolar proton ATPase subunit leads to impaired bone resorption, contributing to osteopetrosis in mutant mice. V-ATPase A1 co-localizes with late endosomal marker Rab7, highlighting V-ATPase A1′s importance in vacuole formation through selective swelling of late endosomes, which is critical for maintaining cellular homeostasis and function.

The description for this antibody indicates it binds to the extracellular domain, but the papers it references points to the endogenous V0 subunit. Can this be clarified please?

Can this antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. We recommend following our recommended protocol, provided below, https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining Alternatively, we do offer V-ATPase A1 (E-8): sc-374475 monoclonal antibody directly conjugated to HRP, and a variety of fluorochromes, if you wish to omit using any secondary detection reagent and potential non-specific binding.

Rated 3 out of 5 by from Weak bandWe tried this antibody for WB in HEK293T cell lysates in 1:1000 dilution. We observed a weak band. Secondary antibody we used was anti-M IgG-680. The results may be improved by using M-IgGκ secondary antibodies.

Rated 5 out of 5 by from IHC detectionWe used this antibody by IHC to detect the expression of V-ATPase A1 in Human colon cancer tissue and normal tissue, V-ATPase was found high expression of V-ATPase A1 in colon cancer tissues, the positive result shows membrane and cytoplasm staining. The results obtained with this antibody are in line with our expectation of the experimental results. We suggest 1:200 dilution for IHC.
