SEE ALSO...

TREX-1 Antibody (E-6) is a mouse monoclonal IgG2a kappa light chain antibody that detects TREX-1 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). TREX-1 (E-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TREX-1, also known as three prime repair exonuclease 1, trophoblast expressed 1, CRV, AGS1, AGS5, DRN3, HERNS, or DNase III, is a crucial member of the exonuclease superfamily and plays a significant role in DNA metabolism and repair. TREX-1 is predominantly expressed in the spleen, liver, thymus, colon, heart, brain, and small intestine, where TREX-1 localizes to the nucleus and functions as a homodimer. TREX-1 is an exonuclease that requires magnesium as a cofactor for activity, demonstrating a preference for double-stranded DNA and operating in the 3′-5′ direction to yield nucleoside 5′-phosphates. Notably, TREX-1 interacts with the SET complex, an endoplasmic reticulum-associated complex, and is involved in granzyme A-mediated apoptosis, highlighting its importance in cellular processes. Mutations or defects in the gene encoding TREX-1 have been linked to several diseases, including systemic lupus erythematosus, chilblain lupus, Aicardi-Goutieres syndrome types 1 and 5, and autosomal dominant retinal vasculopathy with cerebral leukodystrophy, underscoring TREX-1′s critical role in maintaining genomic integrity and cellular health.

We detect an additional band at approximately 25kDa in 293T cells. Is there any information about what this band could be?

Asked by: julia1
Thank you for your question. We do not have any similar data using our TREX-1 Antibody (E-6): sc-271870 in 293T cells. According to Uniprot, there are 3 potential isoforms of TREX-1 predicted at 39, 33 and 32 kDa (http://www.uniprot.org/uniprot/Q9NSU2#sequences). We would recommend performing a secondary antibody control (omit primary antibody) to determine if the band you are observing is a result of non-specific binding of the secondary antibody alone.

When should the TransCruz reagent sc-271870 X be used instead of the standard form sc-271870?

Asked by: jenni
Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.

Rated 5 out of 5 by from Very good antibody for IFI used TREX-1 for IF, the antibody diluted at 1:100 and incubated 3h at room temperature. I used the secondary Goat anti mouse IgG (H+L) Alexa Fluor 568 at dilution 1:500 for 1h. I obtained a specific staining with little background noise.
