TNIK Antibody (C-1) is a mouse monoclonal IgG2b kappa light chain antibody that detects TNIK protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). anti-TNIK antibody (C-1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TNIK, also known as TRAF2 and NCK-interacting protein kinase, is a critical serine/threonine kinase that plays a significant role in cellular signaling pathways, particularly in response to environmental stress. TNIK is composed of 1,360 amino acids and features a protein kinase domain, which is essential for catalytic activity. TNIK is expressed ubiquitously, with highest levels found in brain, heart, and skeletal muscle, indicating importance in various physiological processes. TNIK is involved in phosphorylation of target proteins, a key regulatory mechanism that influences cell division, apoptosis, and cytoskeletal dynamics. TNIK exists in eight isoforms generated through alternative splicing, which may contribute to functional diversity and regulatory roles in different cellular contexts. TNIK monoclonal antibody (C-1) enables researchers to gain valuable insights into complex signaling networks mediated by TNIK and implications in health and disease.

I would like to use Anti-TNIK Antibody (C-1) sc-377215 for western blot. Any suggestion on the use of BSA or Milk for blocking and antibody incubation?

we would like to perform an immunohistochemical staining for TNIK. I found this very interesting monoclonal antibody, offered by you. Now, I was wondering if I should use the pure TNIK Antibody(C-1)(sc-377215) or the HRP-conjugated one?

Thank you for your question. You can use either one. If you use the HRP conjugated form, you do not need to use a secondary antibody, which may be of advantage, but you also can not decide to use the antibody for another application such as immunolfluorescence for example since it's already conjugated. Most customers get the unconjugated form and elect to use a secondary, but either way will work beautifully. Has Santa Cruz done any testing for whether this antibody is specific to TNIK as opposed to the highly related kinases MINK1 and MAP4K4?

Thank you for your question. Yes, this mouse monoclonal antibody, TNIK (C-1): sc-377215, has been validated for specificity using our CRISPR/Cas9 KO Plasmids against the same target gene, TNIK of human origin. This data is shown on our website.

I am using TNIK (C-1): sc-377215 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
