TIP47 Antibody (F-10) is a mouse monoclonal IgG2a kappa light chain antibody that detects TIP47 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). TIP47 (F-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The tail-interacting 47kDa protein (TIP47), also known as human placental tissue protein 17b (PP17b), plays a crucial role in the intracellular transport of lysosomal enzymes by binding to the cytoplasmic domains of cation-dependent and cation-independent mannose 6-phosphate receptors (MPRs), facilitating their movement from endosomes to the Golgi complex. The specific interaction of TIP47 with these receptors is vital for maintaining proper cellular function, as TIP47 ensures that enzymes are correctly sorted and delivered to lysosomes, which are essential for cellular digestion and recycling processes. TIP47 recognizes a phenylalanine/tryptophan signal sequence in the cation-dependent MPR, which is critical for sorting within the endosomal pathway, while TIP47 binding to the cation-independent MPR requires specific cytoplasmic residues for high-affinity interaction. TIP47 competes with the AP-2 clathrin adaptor for binding, highlighting TIP47′s selective role in cargo transport. In clinical studies, TIP47 has been found to be overexpressed in tissue extracts from cervical carcinoma patients, with higher levels observed in dysplastic cells, indicating TIP47′s potential as a biomarker for cancer progression. The gene encoding human TIP47 is located on chromosome 19pter-19p13.3, underscoring TIP47′s significance in both normal physiology and disease states.
