TIP47 Antibody (B-3) is a mouse monoclonal IgG2a kappa light chain antibody that detects TIP47 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). TIP47 (B-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The tail-interacting 47kDa protein (TIP47), also known as human placental tissue protein 17b (PP17b), plays a crucial role in intracellular protein transport by binding to cytoplasmic domains of cation-dependent and cation-independent mannose 6-phosphate receptors (MPRs), facilitating movement from endosomes to Golgi complex. This transport is vital for proper sorting and delivery of lysosomal enzymes, which are essential for cellular homeostasis and degradation of cellular waste. TIP47′s selective binding to specific cargo molecules highlights importance in maintaining efficiency of endosomal pathway. In cation-dependent MPR context, TIP47 recognizes a critical phenylalanine/tryptophan signal sequence necessary for accurate sorting, while for cation-independent MPR, specific cytoplasmic residues are required for high-affinity binding. TIP47 competes with AP-2 clathrin adaptor for binding, indicating significant role in regulation of endosomal trafficking. In clinical studies, TIP47 has been found overexpressed in tissue extracts from cervical carcinoma patients, with higher levels observed in dysplastic cells, suggesting potential as cancer progression biomarker. The gene encoding human TIP47 is located on chromosome 19pter-19p13.3, underscoring genetic significance in human health and disease.

I am using TIP47 (B-3): sc-390981 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Rated 4 out of 5 by from Good antibody for WB and IFThe antibody works well for Western Blot detection of endogenous PLIN3/TIP47 in several human cell lines. Signal is not particularly strong but very clean. Satisfying results in immunofluorescence with both PFA and methanol fixation. Recommended.
