
SHP Antibody (F-12) is a mouse monoclonal IgG2b antibody that detects SHP in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SHP, or short heterodimer partner, is an orphan nuclear receptor that plays a crucial role in regulating various metabolic processes, particularly in the liver, where SHP is predominantly expressed. SHP is unique among nuclear receptors as SHP contains dimerization and ligand-binding domains but lacks the conserved DNA-binding domain, which is essential for traditional receptor function. The absence of this domain allows SHP to act as a negative regulator of other nuclear receptors, including thyroid receptors and estrogen receptors, by inhibiting their DNA binding and competing for coactivator interactions. Additionally, SHP is involved in the modulation of NF-κB transcriptional activity, particularly in macrophages treated with oxidized low-density lipoprotein, highlighting SHP′s importance in inflammatory responses. The regulation of SHP is critical for maintaining cellular homeostasis, especially in the context of bile acid metabolism, where SHP serves as a protective factor against bile acid-induced damage in hepatocytes.
