SAP 61 Antibody (H-3) is a mouse monoclonal IgG2a kappa light chain antibody that detects SAP 61 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SAP 61 (H-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. SAP 61, also known as SF3A3 (splicing factor 3A subunit 3), PRP9, PRPF9, or SF3a60, is a 501 amino acid protein characterized by the presence of one matrin-type zinc finger and is a member of the SF3A3 family. SAP 61 is localized in the nucleus and serves as a crucial subunit of the SF3A splicing factor complex, which is essential for the accurate splicing of pre-mRNA. The SF3A complex, composed of three subunits, facilitates the binding of U2 small nuclear ribonucleoprotein (snRNP) to the branchpoint sequence in pre-mRNA, a critical step in the conversion of the 15S U2 snRNP into the active 17S form that directly participates in splicing events. SAP 61 plays a vital role in this process by interacting with subunit 1 (SAP 114) through its N-terminus while simultaneously binding to the 15S U2 snRNP via its zinc finger domain. This interaction is essential for prespliceosome assembly and overall cell viability, highlighting SAP 61′s importance in gene expression regulation. Additionally, a pseudogene for SAP 61 has been identified on chromosome 20, further emphasizing the complexity of its genetic regulation.
