RAGE Antibody (A11) is a mouse monoclonal IgG2a kappa kappa light chain antibody engineered to target the truncated extracellular domain of the human Receptor for Advanced Glycation End Products (RAGE). RAGE is a type I membrane protein characterized by its distinctive structure, which includes a single V-type immunoglobulin domain followed by two C-type immunoglobulin domains in its extracellular region, a transmembrane domain, and a short cytoplasmic tail. This structural configuration facilitates the binding of various ligands, including advanced glycation end products (AGEs), S100 proteins, and high-mobility group box 1 (HMGB1), enabling RAGE to initiate and propagate diverse intracellular signaling pathways. The extracellular domains of RAGE are essential for ligand recognition and binding, which triggers downstream effects such as the activation of NF-κB and the production of reactive oxygen species, ultimately influencing inflammatory responses, cellular proliferation, and apoptosis. The precise structural features of RAGE also allow for dimerization and interaction with other membrane proteins, enhancing RAGE′s role in cellular communication and signal transduction. Anti-RAGE antibody (A11) is optimized for the detection of natural and recombinant RAGE from mouse, rat, and human origins through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). Additionally, RAGE monoclonal antibody (A11) exhibits reactivity with other species such as bovine, broadening its utility in diverse research settings. By targeting the extracellular domain, anti-RAGE antibody (A11) effectively facilitates the study of RAGE′s structural-functional relationships and its involvement in pathological conditions like diabetes, vascular disorders, and amyloidosis. Can you please provide the amino acid sequence for the A11 Rage antibody. It is extracellular however we would like to know which AA it is associated with SC-80652. We also use the A9 rage which we are aware of the AA sequence. comparing the outcomes.

Dear Santa Cruz, Are you able to tell me the amino acid sequence contained within the 'truncated extracellular domain' that was used as the immunogen for this antibody?

Thank you for your question. RAGE Antibody (A11) is a mouse monoclonal IgG2a raised against a truncated extracellular domain of RAGE of human origin. According to Uniprot accession # Q15109, the extracellular domain corresponds with amino acids 23-342. The exact epitope recognized by this clone has not been determined.

I would like to use RAGE Antibody (A11): sc-80652 for double staining with another mouse monoclonal primary antibody. Is this possible?

Thank you for your question. In cases like this, we recommend using our directly conjugated mouse monoclonal primary antibodies. Currently, sc-80652 is not available directly conjugated, but we have an alternative RAGE antibody (sc-365154) that is available conjugated to HRP, FITC, PE, Alexa Fluor® 488 and Alexa Fluor® 647.
