RAGE Antibody (A-9) is a mouse monoclonal IgG1 kappa light chain antibody designed to recognize the Receptor for Advanced Glycation End Products (RAGE) by targeting a precise epitope mapped between amino acids 23-43 at the N-terminus of human RAGE. RAGE is a crucial type I membrane protein expressed on the surface of endothelial cells, mononuclear phagocytes, and vascular smooth muscle cells, where RAGE functions as a key receptor for advanced glycation end products (AGEs), S100 proteins, and high-mobility group box 1 (HMGB1). The binding of these ligands to RAGE activates several intracellular signaling pathways, including the NF-κB and MAP kinase pathways, which are essential for regulating inflammatory responses, oxidative stress, and cellular proliferation. This functional role of RAGE is significant as RAGE is implicated in the pathogenesis of numerous chronic diseases such as diabetes, atherosclerosis, Alzheimer′s disease, and other inflammatory and vascular disorders. By mediating these processes, RAGE contributes to the progression of vascular dysfunction and amyloidosis, making RAGE a vital target for therapeutic intervention. Anti-RAGE antibody (A-9) is optimized for use in a variety of applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA), demonstrating robust reactivity with RAGE from mouse, rat, and human origins. RAGE is a membrane protein. Why the whole cells were stained with this antibody (Anti-RAGE Antibody (A-9), sc-365154) for immunofluorescence in my experiment?

What is the difference between the Anti-RAGE Antibody (A-9), sc-365154, 200 µg/ml and Anti-RAGE Antibody (A-9): m-IgG Fc BP-HRP Bundle, sc-529316, 200 µg Ab, 10 µg BP? Do the latter include the Secondary Antibody as well?

which dilution should I use for WB? did you try it with Hela cells? Thanks!!

Thank you for your question. We recommend a starting dilution of 1:100 for this antibody for Western Blot. You can see Western blot results with Hela cells above in our published data.

On the figure showing A9 detection of RAGE in various cell types, how much protein was loaded per lane? Also, does this antibody pick up mouse RAGE in Western blots? thanks.

Thank you for your question. We loaded 50 ug whole cell lysate protein. Yes, this antibody is recommended for detection of RAGE of mouse, rat and human origin.

This antibody is noted to be able to be used for FCM. Which concentration of antibody is used for this? I couldn't find it in the protocol sheet. Thanks!

The conjugated primary antibody is recommended and warrantied for flow cytometry (FCM), however we are still gathering data to show on our website. Check back for updates!
