Polycystin-1 Antibody (7E12) is a mouse monoclonal IgG1 kappa light chain antibody that detects Polycystin-1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Polycystin-1 monoclonal antibody (7E12) is raised against amino acids 24-180 corresponding to the N-terminal leucine rich domain of Polycystin-1 of human origin. Polycystin-1 (7E12) antibody is available in both non-conjugated and various conjugated forms, including agarose, phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Polycystin-1 plays a crucial role in regulating cell proliferation and polarity, which is particularly important in autosomal dominant polycystic kidney disease (ADPKD), a condition characterized by cyst formation in kidney tubules and other organs. Mutations in Polycystin-1 are implicated in most ADPKD cases, leading to abnormal cellular behavior within cysts. Polycystin-1 primarily localizes in renal tubular epithelial cells, where Polycystin-1 colocalizes with cell adhesion proteins such as E-cadherin, catenins, vinculin, and paxillin, forming a multiprotein complex essential for maintaining tissue architecture and function. Additionally, Polycystin-1 undergoes post-translational modifications, including tyrosine phosphorylation, which enhances interaction with Polycystin-2 (PKD2) and suggests a role in mediating AP-1 expression. These interactions highlight Polycystin-1′s importance in cell-cell and cell-matrix communication, ultimately influencing cellular responses vital for kidney function and overall homeostasis.

What's the gel percentage used for this polycystin-1 antibody in WB? Also, is there any suitable reference protein or markers?

Thank you for your question. We used a 4-6% gel for this very large protein. These will be very fragile, so handle carefully. Please contact our Technical Service Department for suggestions of suitable high molecular weight proteins that could be used as a reference or molecular weight marker.

Can you provide with a FACS protocol for this antibody (polycystin-1, 7E12) on cell lines?

I recently ordered this mAb conjugated to PE. I'm attempting to use this mAb to quantitate a cell surface antigen - to do that I need to know the ratio of PE molecules per mAb. I.E., do you know if there is one PE bound to each mAb, or two, etc?

Thank you for your question. For PE conjugated antibodies, the ratio is 1:1, so there is 1 PE per antibody molecule.

Is it possible to use the AC conjugated Polycystin-1 Antibody (7E12): sc-130554 AC in both Western Blot and IP?

Thank you for your inquiry. We do not recommend the use of the AC conjugated form in Western blot. The AC conjugated form of the antibody is specifically used for IP. The unconjugated form of the antibody is the best choice when working in multiple applications.
