Pol II Antibody (CTD4H8) is a mouse monoclonal IgG1 kappa light chain antibody that detects the Pol II protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-Pol II antibody (CTD4H8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. RNA polymerase II (Pol II) plays a crucial role in the transcription of protein-coding genes, and Pol II function is vital for gene expression regulation. The enzyme is composed of twelve subunits, with the largest subunit, RPB1 (also known as RPB205), being a 1,840 amino acid protein that features a C2H2-type zinc finger and a C-terminal domain rich in heptapeptide repeats. The structural integrity of RPB1 is essential, as RPB1 not only conveys the catalytic activity of Pol II but also contributes to the formation of the DNA-binding domain, which is critical for the transcription process. The interaction between RPB1 and the other subunits ensures that the transcription machinery is effectively recruited to target promoters, facilitating the initiation of mRNA synthesis. Disruption in Pol II function, particularly through alterations in the largest subunit, can lead to significant impairments in gene expression, underscoring RPB1′s importance in cellular function and development.

With what kind of magnetic beads these Abs work good for ChIP? I tried with Dynabeads Protein G and I didn't get it

Thank you for your question. We have not specifically tested this antibody using magnetic beads, and we do not sell those type of magnetic beads so we can not make a recommendation. This is a mouse monoclonal IgG1, so we would recommend asking the magnetic bead supplier for their recommendation. This specific monoclonal antibody has been validated for ChIP (please refer to the product citations for more details).

According to the info sheet, this antibody was RNAPII Ser5p specific, have you check the specificity against Ser2p/Ser7p or non phopsphorylated RNAPII?

Does the antibody specifically recognizes only Ser5-phosphorylated Pol II CTD? I would like to perform ChIP for total Pol II, as well as Ser5- and Ser2-phosphorylated Pol II. Would you recommend using this antibody as a control for total Pol II?

Rated 3 out of 5 by from Clean but low signalI got a trail size of this antibody and tested it on Murine erythroleukemia cells (MEL). I got clean sharp bands on Western blot, but the signal was very low. I didn't quantify the amount of protein, but the minimum lysate required to see a signal was roughly equivalent to that from 150,000 cells. IMO for Pol II that's a lot!
