PDX-1 Antibody (B-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects PDX-1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PDX-1 (B-11) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The pancreatic duodenal homeobox-1 protein (PDX-1), also known as insulin promoter factor 1, is a crucial 282 amino acid transcription factor that plays a vital role in the development of pancreatic islets and the regulation of insulin gene transcription in beta-cells. PDX-1 is predominantly expressed in the pancreas and duodenum during adulthood, although PDX-1 is present in all cells during early development. PDX-1 function is essential for maintaining glucose homeostasis, as PDX-1 regulates the expression of genes involved in insulin production. PDX-1 interacts with several key proteins, including HNF-3β, HNF-1α, and SP1, which enhance transcriptional activity. Additionally, PDX-1 is modulated by glucagon-like peptide, leading to increased intracellular cAMP levels that activate protein kinase A, further promoting PDX-1 transcription and translocation to the nuclei of beta-cells. Notably, mutations in PDX-1 can result in serious conditions such as maturity-onset diabetes of the young and pancreatic agenesis, highlighting PDX-1′s critical role in metabolic health. PDX-1 is located on human chromosome 13q12.1, underscoring PDX-1′s importance in both developmental biology and disease.

Can PDX-1 (B-11): sc-390792 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, PDX-1 (B-11): sc-390792 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
