I have failed several times with NuPAGE™ Novex™ 4-12% Bis-Tris Protein Gels and transfer with iBlot2 for 10 minute. Do you think this gel is qualified to detect a protein located in 300 kDa? And do you have any suggestion? should I load more sample?

Thank you for your question. We have not specifically used gels and transfer system that you are using. You may want to contact those suppliers for more information about the suitability of their reagents. In our experience with our p300 Antibody (F-4): sc-48343, we recommend using a low-concentration gel and longer transfer times to a PVDF membrane (rather than nitrocellulose) for detection of a larger protein such as p300. We recommend loading 40-60 µg of nuclear extract protein for p300 (e.g., HeLa, MCF7, Jurkat or A-431). For large proteins, transfer out of the gel may be very slow, just as the larger proteins run slowly within the gel during separation. If blotting a large protein, we recommend to run your samples in a low-concentration gel, 8% or less. These gels will be very fragile, so handle carefully. We also recommend to choose wet transfer overnight at 4C instead of semi-dry transfer. The following reference discusses a gel and buffer system that allows transfer of proteins as large as 500 kD: Bolt and Mahoney, High-efficiency blotting of proteins of diverse sizes following sodium dodecyl sulfate– polyacrylamide, gel electrophoresis. Analytical Biochemistry 247, 185–192 (1997).

Can I use this mouse monoclonal antibody for immunohistochemical staining of mouse tissue? Or even double or triple staining on the same samples?

This antibody is currently available conjugated to phycoerythrin (sc-48343 PE), fluorescein (sc-48343 FITC), Alexa Fluor® 488 (sc-48343 AF488) or Alexa Fluor® 647 (sc-48343 AF647). Rated 3 out of 5 by from Band at expected height I have tested this antibody in primary breast and lung cell lysates, using Tris/Tricine gradient precast gels for high molecular weight proteins. I tested the antibody 1:100 in 5% BSA, and used an ECL-compatible secondary antibody. There was clear signal around 300 kDa, although there was some background (perhaps changing blocking buffer may solve this).
