Selectins, also designated CD62 antigens, comprise a family of carbohydrate-binding proteins involved in mediating cellular interactions with leukocytes. L-Selectin (also designated LECAM-1 or CD62L) is expressed on the majority of B and naive T cells and on most monocytes, neutrophils and eosinophils. L-Selectin interacts with specific carbohydrates expressed by activated endothelial cells. P-Selectin (also designated GMP-140 or CD62P), expressed on activated platelets and endothelial cells, and E-Selectin (also designated ELMA-1 or CD62E), expressed on endothelial cells, exhibit overlapping ligand specificities. Both recognize sialyl-Le (x) as a ligand and bind to specific carbohydrates on neutrophils and monocytes.

Regarding my question about another antibody, I'm not looking for a secondary antibody, but for another P-selectin antibody that could be used as the detector in ELISA that would bind at a spatially distinct epitope. Can you recommend one? Thanks
Asked by: tracey01
Thank you for your question. We do not characterize pairs of monoclonal antibodies for use in sandwich ELISA. Many of our antibodies are recommended as detection antibodies in direct or indirect ELISA, however we cannot recommend a matching capture antibody. Please contact Technical Service by phone, (800)-457-3801 option 2, email <[email protected]>, or by live chat directly on our website, www.scbt.com if you have any further questions.

Do you have any data on performance in ELISA? If the antibody is known or predicted to work in ELISA, can you recommend a second antibody to use in the ELISA pair?
Asked by: tracey01
Thank you for your question. We have not yet tested this specific mouse monoclonal, P-Selectin Antibody (1 E 3): sc-19672, for ELISA so we do not have any specific recommendations we can offer at this time for this research application. However, the antibody has been tested in Western blot, immunohistochemistry and flow cytometry so we believe it should also work for ELISA.
P-Selectin Antibody (1 E 3) is a mouse monoclonal IgG2a, so for a secondary antibody you would want to use an anti-mouse IgG secondary antibody conjugated to either alkaline phosphatase (AP) or horseradish peroxidase (HRP), depending on what detection method being used. We would recommend our mouse IgG binding protein conjugated to HRP, m-IgG kappa BP-HRP: sc-516102. A link to the product description is provided below for your reference.

Can P-Selectin (1 E 3): sc-19672 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?
Asked by: cjMara
Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, P-Selectin (1 E 3): sc-19672 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Dear Mr. or Ms.
We would like to know if this CD62P antibody cross reacts with canine platelets.
I look forward to hearing from you.
Asked by: Peres Badial
Thank you for your question. We do not know whether or not this antibody will detect canine. P-Selectin Antibody (1 E 3) is a mouse monoclonal raised against fixed thrombin-activated human platelets. The exact epitope recognized by this antibody has not been determined so we are unable to try to predict cross-reactivity based on sequence analysis. This would have to be determined experimentally.
