SEE ALSO...

p-RKIP (80.Ser 153) is a mouse monoclonal IgG2a kappa light chain antibody that detects Ser 153 phosphorylated RKIP of human origin by western blotting (WB) and enzyme-linked immunosorbent assay (ELISA). anti-p-RKIP antibody (80.Ser 153) is available in both conjugated and non-conjugated forms, providing flexibility for various experimental needs. Raf kinase inhibitory protein (RKIP), also known as PEBP, plays a crucial role in modulating the Raf/MAPK signaling pathway, which is vital for regulating cell growth, differentiation, and survival. Phosphorylation of RKIP at Serine 153 influences RKIP′s ability to interact with other proteins in the signaling cascade. When protein kinase C phosphorylates RKIP in response to stimuli such as phorbol esters or epidermal growth factor, RKIP undergoes a conformational change that allows dissociation from Raf-1 and association with G protein-coupled receptor kinase 2 (GRK2). This interaction effectively inhibits GRK2 activity, thereby impacting downstream signaling events. RKIP′s ability to modulate these pathways is essential for tumor suppressor function, inhibiting NFkB activation in response to inflammatory cytokines like TNF-α and interleukin-1 beta, ultimately influencing cancer progression and metastasis.

Gent.mi,  How did you check the specificity of this antibody?  We are testing it by ELISA but It seems that It recognizes both native and p-153 RKIP with the same affinity...It should not happen.

Asked by: Marrabio10
Thank you for your question. p-RKIP Antibody (80.Ser 153): sc-135779 has been validated by WB and ELISA for specificity against Ser 153 phosphorylated RKIP of human origin. Please contact our Technical Service Department for help with troubleshooting the antibody if you have results that indicate the antibody is non-specifically recognizing the non-phosphorylated antigen. Please call our European Support office toll-free at 00800 4573 8000, email europe@scbt.com, or contact us by live chat.

How much sample do you recommend loading on the gel for Western blot detection using p-RKIP (80.Ser 153): sc-135779?

Asked by: jenniferc
We recommend loading 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane. A link to our complete Western blotting protocol is provided here for your reference. https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting

Rated 5 out of 5 by from Potent and very specific antibodyThis monoclonal antibody was independently tested by Kinexus Bioinformatics Corporation and is recommended for Western blotting. There was very strong detection of RKIP in 30 minute anisomycin-treated human HeLa cells on Western blots, and no significant cross-reactivities on these blots with any off-target proteins.
