Raf kinase inhibitory protein (RKIP, PEBP) is a modulator of the Raf/MAPK signaling cascade and a suppressor of metastatic cancer. RKIP inhibits MAPK by preventing association of Raf-1 and p21-activated kinase (PAK), and blocking phosphorylation of the Raf-1 kinase domain by PAK and Src kinases. After G protein receptor stimulation, RKIP can dissociate Raf-1 and associate with GRK2, thereby blocking GRK2 activity. This switch is triggered by protein kinase C (PKC)-dependent phosphorylation of the RKIP on Serine 153. RKIP Serine 153 phosphorylation by PKC in response to phorbol ester or epidermal growth factor causes release of RKIP from Raf-1. RKIP antagonizes the signal transduction pathways that mediate the activation of NFkB in response to stimulation with TNF-a or interleukin-1 b.

How did you check the specificity of this antibody? We are testing it by ELISA but It seems that It recognizes both native and p-153 RKIP with the same affinity...It should not happen.

Thank you for your question. p-RKIP Antibody (80.Ser 153): sc-135779 has been validated by WB and ELISA for specificity against Ser 153 phosphorylated RKIP of human origin. How much sample do you recommend loading on the gel for Western blot detection using p-RKIP (80.Ser 153): sc-135779?

We recommend loading 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane. A link to our complete Western blotting protocol is provided here for your reference.
