Orexin-A Antibody (KK09) is a mouse monoclonal IgG1 antibody designed to detect Orexin-A across mouse, rat, and human species using western blotting (WB), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). Anti-Orexin-A antibody (KK09) targets a synthetic peptide corresponding to amino acids 34-66 within an internal region of human Orexin-A, ensuring selective binding without cross-reactivity to Orexin-B. Orexin-A, also referred to as Hypocretin-1, HCRT, and PPOX, is a critical hypothalamic neuropeptide derived from the precursor prepro-orexin, playing an essential role in maintaining homeostasis by regulating temperature, thirst, and hunger. Orexin-A exerts effects by binding to Orexin receptor-1 and Orexin receptor-2, which are G protein-coupled receptors involved in the integration of the endocrine and nervous systems. Orexin-A is predominantly localized to neurons in the lateral hypothalamus, known as the "feeding center," and expression is upregulated during fasting, highlighting importance in the central feedback mechanisms that govern feeding behavior. Additionally, Orexin-A enhances the release of neurotransmitters such as GABA and glutamate, facilitating synaptic activities within the hypothalamic region. Orexin-A monoclonal antibody (KK09) is an invaluable tool for researchers studying the physiological and pathological roles of orexins in energy balance, sleep regulation, and neurodegenerative diseases.
